Modulation of anti-inflammatory cholinergic pathway in the liver of the offspring by maternal consumption of high-fat diet

Abstract

Maternal obesity is related to the development of metabolic disorders in the offspring. Exposure to fat intake during pregnancy and lactation can lead to obesity and increased inflammatory markers in adulthood. An event that has been widely investigated is the contribution of maternal nutritional challenges during pregnancy and lactation, for metabolic programming and development of metabolic disorders. The inflammatory process triggered by maternal consumption of high-fat diet seems to be triggered by activation of the TLR4/NFkB. From this one protein is activated protein cascade leading to production of cytokines, which stimulate its receptors feeding the inflammatory response. The fine control of this response, called the inflammatory reflex, which avoids the increased production of TNF and IL1² can promote damage to the body. The cholinergic anti-inflammatory pathway is activated via the vagus nerve and by the alpha7 subunit of the nicotinic acetylcholine receptors (alpha7nAChR) reduces activation of NF-kB and consequently decreases the production of TNF and intensity of the immune response. The inhibition of these receptors or denervation of the efferent pathway causes intense production of cytokines in response to LPS. In this sense, studies with experimental models have shown that the offspring of obese mothers presents an exaggerated inflammatory response after a challenge with LPS or high fat diet. Thus, it is possible that these animals exhibit control of fine-tuning the inflammatory response affected by maternal consumption of high-fat diet during pregnancy. The liver is an organ that presents the Kupffer cells which are resident macrophages and that participate in inflammatory processes associated with harmful fatty liver, and obesity. Thus, the central aim of the project is to evaluate the anti-inflamatória cholinergic pathway in the offspring of obese mothers and try to establish a relationship between liver damage in offspring caused by maternal consumption of high fat diet and damage in the components of cholinergic pathway.
