Abstract

The objective of this work is to study the kinetics of cellobiose enzymatic hydrolysis and assess the mechanism and the degree of inhibition of glucose on ²-glucosidase from Aspergillus niger (Novozyme 188). The experimentally measured initial velocity of the reaction will be used to determine the kinetics of inhibition by product in the following conditions: cellobiose and glucose concentrations of 1-20 g / L and 2-20 g / l, respectively. The experiments will be performed at pH 4.8, temperature 50°C and enzyme load fixed at 38.5 CBU / mL solution. The kinetic parameters (Ki, Km and Vmax) will be determined using the reciprocal Lineweaver-Burk graphics. (AU)
