Determination of soluble CD163 and cytokines in plasma of pregnant women with preeclampsia

Abstract

Preeclampsia (PE) is a pregnancy-specific syndrome that manifests from the 20th. week of pregnancy and is characterized by intense inflammatory response. Monocytes from peripheral blood of pregnant women with pre-eclampsia are endogenously activated and secrete high levels of inflammatory cytokines and free radicals. The CD163 molecule is considered a specific marker of monocytes / macrophages and is released in plasma under pathological conditions. Elevated levels of soluble CD163 (sCD163) in plasma is considered a marker of monocytes / macrophages activation and correlate with inflammatory diseases activity and progression. This project aims to determine whether there is an association between plasma levels of sCD163 and pro-and anti-inflammatory cytokines with disease severity in pregnant women with PE. Eighty-four pregnant women, 28 normotensive, 28 patients with mild PE and 28 patients with severe PE, matched for gestational age were included, and a group of 28 healthy non-pregnant women of the same age of pregnant women will be evaluated to establish the comparison between the groups. Peripheral blood of pregnant women and healthy women will be collected to determine sCD163, IL-1b, IL-6, IL-10, TGF-b1 and TNF-a by enzyme immunoassay. This project intends to determine whether this soluble receptor in plasma and its association with cytokines may be considered a marker of monocyte activation, severity and prognosis of the disease.
