ERK 1/2 protein expression in response to aldosterone treatment in rat kidney

Abstract

Studies show that extracellular signal-regulated kinases (ERK1 and ERK2) are involved in aldosterone nongenomic mechanism in the kidney, participating of ionic transporters regulation along the nephron. Considering that the hormonal action mechanism isn´t totally clear, but it´s already known the importance of ERK1/2 to start the aldosterone fast action on the kidney, the aim of this work it´s to study the acute effects of aldosterone on the renal's rats total and phosporilated ERK1/2 expression and the GR and MR involvement on this process. For that, the aldosterone will be administrated into the rat's peritoneum (Wistar)and after 30 minutes, the animals will be anaesthetized and the kidneys will be taken off to measure total and phosphorilated ERK1/2 protein expression's on the cortical and medullar region, with the western blot technique. Also will be evaluated the eplerenone's (MR's receptor inhibitor) and RU486's (GR's receptor inhibitor) effects on the phosporilated ERK1/2 expression.
