Abstract

The use of stem cells originating from pulp tissue has frequently been related among tissue regeneration techniques. However, the characterization of these cells and which population (pulp cells from primary or permanent teeth) would be more efficient for the various types of therapies has not yet been described in a detailed manner. Therefore, the aim of the present study will be to expand primary culture of pulp cells obtained from exfoliated human primary teeth and 3rd molars, using different techniques, and compare the characteristics and quantity of stem cells in each model, by means of specific tests. For the study, the pulp tissue of 10 exfoliated human primary teeth and 10 third molars will be obtained and divided into two parts, and the cells will be isolated by two types of techniques (explant and enzymatic digestion). After culture expansion, from the 2nd passage, specific tests will be performed for the characterization and identification of stem cells. Immunophenotyping will be performed by means of immunofluorescence and flow cytometry tests, using specific antibodies for marking stem cells (nestina, vimentina, SH2, SH3 and Oct3/4). In addition, kariotyping of cells will be performed, with the object of confirming the absence of raw chromosomal alterations during culture. A quantitative analysis will be performed, with images acquired by immunofluorescence and kariotyping tests. Whereas, for flow cytometry, the data will be evaluated as regards their distribution; should there be normal distribution, the fixed two-way analysis of variance (cell type and number of passage) will be applied, (type of cell and passage number) complemented by the Tukey test, if necessary. Should the distribution not be normal, the non-parametric tests such as those of Kruskal-Wallis and Mann-Whitney will be used. The level of significance will be 5%. (AU)
