Efect of BAY 41 22-72, soluble guanylate cyclase agonist, in human T cells and Jurkat cells.

Abstract

Primary immunodeficiencies (PID) are disorders that predispose to recurrent infections, autoimmunity, allergy, cancer, and other immune dysregulation. These diseases may involve elements from innate, as mononuclear phagocytes, polymorphonuclear cells, NK cells and complement proteins, or acquired immunity, such as T and B lymphocytes. Although all efforts invested in PID research, the progress in diagnosis and treatment of these diseases still a challenge. Our group has contributed to study of new therapies or drugs with therapeutic potential for PID patients. Recently, we demonstrated that BAY 41-2272, a soluble guanylate cyclase (sGC) agonist, proved to be effective in modulate immune cells, leading to infection control. In T cell, has been shown that CG activation by low levels of NO increases cGMP levels, selectively inducing IL-12 receptor beta2 (IL-12R²2) expression. It was also shown that increasing concentration of cGMP and, consequently, higher intracellular calcium influx induces IL-4 production. Considering these facts, BAY 41-2272, and its signaling pathway, presents a great potential for activation of T lymphocytes. Thus, we propose to investigate the effects of in vitro treatment with BAY 41-2272 on Jurkat cell line and CD3+ T cells from healthy donors. We intend to evaluate BAY 41-2272 effects through assessing cytokine production, immunophenotyping, cell proliferation, cell viability studies signaling pathways and co-cultured with monocytes.
