Analysis of the active MMP-2 and MMP-9 production by malignant epithelial cells and the influence of the invasion model assay in cell culture

Abstract

This project is part of a major project that will study in in vitro assay the influence of benign myoepithelial cells of pleomorphic adenoma in MMPs -1, -2, -8, -9 and -13 production by malignant epithelial cells and their relationship with invasion index. In the present project, the production of active MMP-2 and MMP-9 by malignant epithelial cells of squamous cell carcinoma and ductal carcinoma, and the influence of the invasion assay will be analyzed in cell culture. For this, malignant epithelial cells will be cultured in 24 wells plates and in transwell chambers with 8¼m pores membrane coated with matrigel adapted in these plastes as well. The conditioned medium will be collected after 96h. The presence of active MMP-2 and MMP-9 in these medium will be investigated and compared by the proteolytic activity using gelatin zimography.
