Abstract

The oral cancer represents 7% of human cancers, where the squamous cell carcinoma (SCC) is the most common type. The transition from normal mucosa to invasive carcinoma has a complex multistep and multifactorial etiology. The mucosal changes that precede the occurrence of a malignant lesion have a great interest in this context, where the oral potentially malignant lesions (PMLs) are the targets of several studies. Matrix metalloproteinases (MMPs) are calcium-dependent endopeptidases responsible for the degradation of the extracellular matrix. The MMPs may be related to the degradation of the basal lamina and possible progression of epithelial dysplasia for SCC and subsequent invasion of adjacent tissue. Some studies have shown a possible interaction between the transcription factors Runx1 and ETV5 with a series of events intra- and extra-cellular, culminating in neoplastic progression and invasion in skin SCC and endometrial carcinomas. Among these interactions it was observed that the Runx1 and ETV5 stimulate the production of MMPs 2 and 9. Hence, it is suspected that this interaction may be related to malignant transformation of PMLs, as well as the progression of oral SCC. This project aims to evaluate and correlate the expression of transcription factors Runx1 and ETV5 with the expression and activity of MMPs 2 and 9 in oral PML and SCC. The study will be performed on frozen tissue samples derived from PML, SCC and normal oral mucosa, using the techniques of immunohistochemistry, western blot, zymography and immunofluorescence with triple labeling. All results will be correlated with the clinical and pathological data of each sample will be analyzed by statistical methods. (AU)
