Abstract The aim of this study is to evaluate the MMPs activity (MMPA) of primary and permanent dentin over time as well as the influence of the treatment with EDC on the inhibition of these proteases and in vivo degradation of the adhesive interface. In the Experiment 1 dentin beams will be obtained from primary and permanent molars and completely demineralized. MMPA, initial modulus of elasticity (E) and dry mass will be determined for each group (primary and permanent). The beams will be stored in artificial saliva for up to 2 months and the new E and dry mass will be reassessed after 7 days, 1 month and 2 months of storage, as well as the release of ITCP and CTX. For Experiment 2, third molars will be used to produce dentin beams. After their demineralization the initial E, MMPA and dry mass will be determined. Then, the beams will be treated with the following solutions for 60s: water (control), 0.1M, 0.3M, 0.5M, 1M or 2M EDC or 5% glutaraldehyde. After the tratment, E and MMPA will be further evaluated and the specimens will be stored in artificial saliva. The E, dry mass and MMPA will be remeasured after 7 days, 1 month and 2 months. Finally, in Experiment 3, ten patients with caries in primary molars will be selected. After caries removal and acid etching the dentin will be treated with water (control) or 0.5M EDC for 60s prior to application of the adhesive system. The teeth will be restored and after exfoliation will be collected for analysis of the adhesive interface by TEM. Data set for MMPA, E and dry mass will be analyzed for normality and homocedasticity to guide the selection of the statistical tests (p>0.05). TEM images will be analyzed descriptively. | |
