Effect of progesterone on the expression of genes related to follicular development in granulosa cells of Nellore heifers

Abstract

High progesterone concentration during synchronization protocol leads to a small follicular development with smaller pre ovulatory estradiol concentration and progesterone post ovulation in Nellore heifers. This occur probably due less LH pulsatility that affect negatively follicular development post divergence. Recent experiment showed the effect of LH on LH receptors in granulose cells. This present study aims evaluate if progesterone concentration interferes on expression of LH receptors on granulose cells from Nellore heifers. Will be used 52 Nellore heifers that will be pre synchronized (d-17: EB + progesterone device CIDR® ; D-10: PGF2±; D-8: ECP and CIDR removal). On D0 all follicles will be ablated and heifers replaced in 6 groups: CIDR 1 (new) x CIDR 3 (Previously used for 18 days) in three periods; Treated with CIDR for 5 days and slaughtered on day 5; Treated with CIDR for 6 days and slaughtered on day 6 (longer P4 exposition ; Treated with CIDR for 5 days and slaughtered on day 6 (exposed to a period without exogenous P4= longer proestrus). Is expected that the expression of LH receptors on granulose cells will be determined by P4 concentration that the follicle was exposed (“P4 = ‘competency) and length from the follicle emergency (‘length = ‘competency).
