Abstract

Studies have revelead that membrane amniotic human, feline and canine are a good source of mesenquimal stem cells (Msc). Your getting are free of ethical conflicts because the amnion is commonly discarded after birth. Due of innovative features of stem cell use on regenerative medicine, many tests are been performed to answer questions about possible complications of his use such as: infections at the time of application, tissue deterioration and especially about the carcinogenic potential of cell lines. Teratogenic tests were conducted in humans and in cats, and the result was negative for both species, the same test was performed on dogs with positive results for teratoma condemning your use. The purpose of this study is to trace a profile molecular and teratogenic tests thorough comparison between the amniotic membrane of the canine and feline, since the experiments were favorable in this species, aiming discover at which point they differ which may justify the results obtained in previous studies with amnion canine. For the analysis of the cells will be used amniotic membrane obtained from neonates born by elective cesarean or castrations pregnant bitches aged 30-45 days of the FZEA Veterinary Hospital (UDHC) and neutering campaigns in the city of Pirassununga. The amnion is separated manually from other fetal membranes and placed in petri dishes. It will be washed in a solution of PBS-L, manually homogenized, subjected to enzymatic digestion and then cultured in culture medium for mesenchymal cells. After the establishment of the culture will be carried RT-PCR analysis, immunohistochemistry and flow cytometry for the identification of mesenchymal markers CD90, CD73 and CD105, hematopoietic markers as CD45 and CD34, tumor markers as vimentin, nestin, p-53, c-Myc, and CD30 beyond carcinogenic testing in immunosuppresseds mices.
