Analysis of transcriptomes of Xeroderma pigmentosum cells of patients after irradiation with UVA light

Abstract

The Sequencing of Transcripts (RNA-Seq) of cultured primary fibroblasts irradiated by UVA light is being developed by the DNA Repair Laboratory, at the Department of Microbiology of ICB/USP. This study is developed with fibroblasts of Brazilian individuals clinically diagnosed with Xeroderma Pigmentosum Syndrome (XP), which have different types of defects in repair and/or tolerance of DNA damage. The transcriptome sequences will be analyzed to look for information on expression patterns and differences between transcribed genes. To this end, it is required the interpretation of a large amount of data obtained in the sequencing, as well as the processing of this data through specialized software/pipelines process. The work proposed here is primarily engaged in performing analyzes of transcriptomes of cells (primary cultures) XP-V and normal, non-irradiated and irradiated with UVA light, in the quest to understand the effect of these wavelengths in gene expression in cells with deficiency in translesion synthesis of DNA. For this, evaluation, discussion and construction of protocols and bioinformatics tools will be performed to identify differential expression levels as well as alternative splicing induced by UVA irradiation in these cells.
