Abstract

In mammals, oocytes are removed from the follicular environment, no longer have the signal that maintains meiotic block and restart meiosis. In bitches, although the oocyte meiosis can resume spontaneously in vitro maturation rates are very low and the percentage of oocytes that remain VG is high. Several studies have attempted to understand the cell cycle regulation in mammalian oocytes, especially in regulating a number of protein kinases and phosphatases which control meiosis in order to elucidate what remains blocked meiosis. Thus, this project aims to consolidate and extend the experimental study and applied, with emphasis on cell cycle control of canine oocytes and its association with oocyte competence in this species through molecular techniques refined to identify the expression of their target biomarkers. The ovaries will be collected from bitches considered otherwise healthy on clinical examination and submitted to ovariohysterectomy (OSH) elective. The oocytes will be selected for in vitro maturation (IVM) and assessed at 0h, 24h, 48h and 72h. The technique of proteomics will be used to identify and quantify the expression of proteins involved in cell cycle control and technique of immunofluorescence to assess the location of these proteins during the different stages of IVM.
