Abstract

Congenital diaphragmatic hernia is a defect in the development of the diaphragm that occurs in 1:2500 newborns with high fetal and neonatal mortality secondary to pulmonary hypoplasia and hypertension. This defect can be experimentally created in rabbit fetuses at 25 days of gestation with 50% of fetal survival. Histologically, the lungs in CDH show disturbed alveolarization and vascularization leading to respiratory failure and difficulties in ventilating the newborn. Besides, the pulmonary hypertension increases the right ventricular, which has implications on follow up progress and survival. Our objective is to investigate the cardiac function and myocardial contractility immediately after delivery in the surgical model of CDH. We will study the in vitro contractility of the myocardium from the left and right ventricles and compare it to control fetuses. Following, we will study the effect of vasoactive drugs on the myocardium and its possible implications to pulmonary hypertension and myocardial function in CDH. (AU)
