Impact of systemic treatment with curcumin on the progression of experimental periodontitis in the presence of rheumatoid arthritis

Abstract

This study aims to evaluate the effect of systemic treatment with curcumin on the progression of periodontitis-induced experimentally in animals with rheumatoid arthritis (RA). Additionally, we will also investigate the impact of curcumin in control of RA in animals with induced periodontal disease. To this, 24 males and adult rats (Rattus norvegicus, Albinus, Wistar) will be randomly divided into the following groups: 1 - + experimental arthritis experimental periodontitis + placebo (RA + PE + PL) and 2 - experimental arthritis experimental periodontitis + + curcumin (RA + PE + C). Rheumatoid arthritis (RA) will be induced by two immunizations with type II collagen and two applications of Freund's complete adjuvant (FCA). The first dose of collagen will be administrated on day -21 and the second dose on day -14. On day -7 two applications of CFA will be held. On day 0, the PE will be induced by placing a ligature of silk thread in the groove of the mandibular first molars. Weekly tests (evaluation of paw edema and analgesic test) will be held until the end of the experiment to confirm the induction of RA. After this period, the animals are sacrificed and the specimens will be processed for morphometric analysis of alveolar bone loss. The gum tissue will be collected for quantification of IL-17 by Luminex assay / Magpix. Data will be analyzed statistically using the Student t test or Mean Whitney, after normality test (± = 5%).
