Hepcidin quantification and measurement of serum iron in dogs with pyometra

Abstract

The discovery of hepcidin is one of the great revolutions in hematology, being mainly associated with iron homeostasis . It is a peptide of 25 amino acids , synthesized primarily in the liver ( Nemeth and Ganz , 2006) , which has its synthesis linked to infections and inflammations . Acts as regulator of the efflux of iron from tissues into plasma ( Nemeth et al , 2003; . Kemna et al , 2005. ) , being responsible for hypoferremia with subsequent gradual installation of a non- regenerative anemia.The aim of this study comprises quantify and evaluate the behavior of hepcidin in bitches with cystic endometrial hyperplasia ( HEC ) / pyometra as well as perform the measurement of serum iron and evaluate their behavior compared to that of hepcidin .Bitches with pyometra assisted at the Veterinary Hospital of Obstetrics FMVZ - USP and forwarded to perform ovariohysterectomy ( OSH) will be included in the study . The intended sample size is 30 cases of pyometra and 30 other healthy dogs that will compose the control group .Two blood samples will be collected, the first held in the first visit of the Service of Obstetrics and second in return for clinical surgical reassessment ( 7-10 days) , totaling 90 samples to be process .Blood concentrations of hepcidin are quantified by an enzyme immunoassay (ELISA) using specific commercial kit . To review the ferremia , measurement of serum iron and its total binding capacity through the colorimetric method is performed using the commercial kit from Randox ® brand .Data will be statistical evaluated using THE program Sigma Stat . Possible differences between groups will be analyzed with the Wilcoxon paired test . The ± level of significance is 5% (0.05).
