Abstract

Silver has been largely incorporated in the nanomaterials due to its broad-spectrum antimicrobial action. Currently, the silver nanoparticles (AgNP) are being released into the environment in amounts that are becoming an environmental concern. Studies about AgNP discuss whether the toxicity of this nanomaterial is due the ionic silver dissolution or if the nanoparticles themselves are the responsible by the high toxicity. The methodological difficulty in the Ag determination in biological materials and in low concentrations, mainly in saline samples (marine environment), is a limiting factor for toxicological studies with this metal. In this work, will be developed and evaluated a method for the determination of silver in the Brazilian marine amphipod, Parhyale hawaiensis, exposed to nanoparticles of this metal through feeding. Besides that, copper concentration, which is naturally present in the hemolymph of these animals, will be used as an intern standard. Parhyale hawaiensis individuals will be exposed to AgNP incorporated in the food and, after that, dissection and separation of hemolymph and organs/tissues of the exposed individuals will be done. The procedures of biological material digestion and interferences elimination will be evaluated, aiming the determination by atomic spectrometry (absorption and/or emission). Concentrations of Ag and Cu and the Ag distribution in the different fractions of the biological material will be determined. Also, the seawater will be analyzed. From the results, it is expected to obtain information about the absorption, destination and storage of silver in the amphipod, providing data for the development of biomarkers of exposure. (AU)
