Study of the pathophysiology of acute inflammatory reaction during infectious aerocistite by Aeromonas hydrophila in Nile Tilapia, Oreochromis niloticus, alloxan-diabetic

Abstract

Given the significant results observed in the evaluation of acute inflammatory reaction in diabetic tilapia with impaired innate immune response, particularly in the suppression of phagocytes into the inflamed focus, associated with changes in blood leukocyte populations primarily in lymphocytes and neutrophils, and histopathological changes serum biochemical- an increase of lipids, proteins and glucose (1st phase of the study, dissertation, process 2011/16945-9 FAPESP), this study proposes to investigate other mechanisms involved in the pathophysiology of acute inflammatory reaction in alloxan-diabetic tilapia through analyzes: serum levels of acute phase proteins; serum concentrations of IL- 1², IL- 6 and TNF- ±; Tissue expression of IGF- 1, IL- 1² and TNF- ± receptors; determining circulating levels of insulin and cortisol; Serum levels of sodium, calcium and potassium; Determination of Serum lysozyme. To this serum and tissues of tilapia with 189 acute infectious aerocistite by A. hydrophyla (process 2011/16945-9 FAPESP) will be used, constituting 9 treatments: Untreated and not inoculated (negative control); Untreated and inoculated; Treated with insulin (INS) and inoculated; + INS treated with alloxan (ALOX) and inoculated; Treated with insulin (INS) + dexamethasone (DEXA) and inoculated; ALOX treated and inoculated; Treated and inoculated ALOX + DEXA; DEXA treated and inoculated; Treated with INS + + ALOX DEXA and inoculated. This step is also conducted an experimental trial with 30 alloxan diabetic tilapia for determining body weights (as appended pilot study), comprising three groups (n = 10): non-diabetic and treated with saline (G0); Diabetic and treated with saline (G1); Diabetic and treated INS (G2). The global values of body mass (g), bone mineral density (g/cm2), density of fat mass (%) and density of lean mass (g) will be analyzed. However, the correlation between studies: Acute phase proteins; serum cytokines; structural cell immunohistochemistry; determination of body mass; circulating levels of insulin, cortisol, lysozyme and electrolytes, associated to the results observed in hematological studies, biochemical, histopathological, and somatic cell in the inflammatory exudate (obtained in the 1st phase of the study, proc. FAPESP 2011/16945-9), provide subsidies important in understanding and characterization of events in the pathophysiology of acute inflammatory reaction in alloxan-diabetic tilapia and endocrine, metabolic and immunological involvement of the insulin effect in organic and glucocorticoid response of the teleost fish. Thus, this study has two major goals: to disclose important pathophysiological mechanisms involved in acute inflammation of diabetic tilapia and prove the importance of this experimental model teleost fishes for studies and advances in research into diabetes mellitus.
