Abstract

The use of cryopreserved equine semen has grown in recent years due to the ease on transportation and storage of genetic material from stallions for a long period of time. However, there is great influence of semen processing on stallion sperm quality. Among other factors, it is known that the cryopreservation process involves oxidative stress on sperm cells, and the addition of antioxidants to cooling and freezing semen extenders may help protect sperm against damage induced by Reactive Oxygen Species (ROS). Among the diverse antioxidants, Butylated Hydroxytoluene (BHT), a synthetic analogue of vitamin E, may improve sperm viability once included in cooling and freezing extenders. This experiment aims to evaluate the effect of BHT on extender on survival, motility, plasma membrane integrity and mitochondrial activity of equine spermatozoa before and after cryopreservation - freezing and cooling, and to evaluate the ability of BHT in protecting the sperm from the toxic effects of ROS. The inclusion of this antioxidant in cooling and freezing extenders might improve sperm viability, not only for stallions considered good coolers and good freezers, but specially for those animals classified as poor coolers and poor freezers.
