Abstract

The Lambari-da-Mata-Atlântica, Deuterodon iguape, is an endemic fish of the Ribeira de Iguape hydrographic basin and presents large potential for captive breeding, however, technological aspects related to reproductive processes need to be studied. Thus, the aim will be to standardize and to evaluate the computerized sperm parameters using open source software ImajeJ/plugin CASA. Will be collected the sperm of 20 mature males hormonally induced (single dose of 2.5 mg of CHCE kg-1). After 12 hours of induction, the animals will be subjected to semen collection by manual extrusion in the cephalo-caudal direction. The sperm will be activated with distilled water and evaluated on a trinocular microscope, fitted with image capture system. The videos will be recorded throughout the duration of sperm movement. After recording, the video will be edited and evaluated in the ImageJ software/plugin CASA. The settings of the program will be standardized according to sperm size and movement through of the three phases: 1) standardization of CASA plugin settings; 2) frame rate used to perform the analysis and; 3) motility between 10 and 60s post-activation with analysis every 5s. The results will provide of basis for future studies of artificial reproduction of the specie.
