Abstract

In Brazil, most native species produced in captivity require artificial reproduction to reproduce and, generally, the greatest concern is regarding the quality of female gametes, the oocytes, with the quality of male gametes taking second place. However, if seminal quality is not adequate, fertilization of the oocytes will be inefficient. In this sense, seminal quality is a fundamental factor for fish production. Therefore, knowing the methodologies for seminal analysis is a basic factor both at the research level and at the producer level. More efficient methodologies for seminal analysis are currently available, such as CASA - Computer Assisted Sperm Analysis, which allows a more precise evaluation of a greater quantity of kinetic and morphological parameters of fish sperm. Its use has been crucial for a more assertive selection of breeders, in the development and optimization of gamete cryopreservation protocols, thus improving the level of fertilization. However, due to the great variability of fish species, a great variability of seminal aspects, requiring technical and biological knowledge to make adjustments to the system that will ensure the accuracy of the results. This project will be developed at the Research Institute of Fish Culture and Hydrobiology, Faculty of Fisheries and Protection of Waters, University of South Bohemia in ¿eské Bud¿jovice, located in the Czech Republic, in the city of Voldnany. I will be under the guidance of Dr. Borys Dzyuba. My objectives are: to expand my knowledge of the CASA system for characterizing fish semen, within the various possible approaches, aiming to improve the quality of the desired results for my scientific initiation project and, also, to learn seminal cryopreservation protocols.
