Abstract

Head and neck cancer(CCP) is the fifth most common type of cancer and the survival rate has not changed in recent years. Chemotherapy treatment for the disease has shown unfavorable results due to tumor resistance. This fact is possibly due to the existence of a small subpopulation of cells in these tumors with properties of stem cells, called tumor stem cells (CTT), responsible for sustaining tumor growth by tumor resistance to conventional therapies and recurrence of disease. Therefore, the objectives of the study is to identify the presence of CTT in CCP and cell lines, to verify the effectiveness of chemotherapy in different conditions of exposure time, to analyze mRNA expression of genes related to the presence of CTT and the association of mRNA expression at the level of aggressiveness of the tumor. First the CTT will be identified by flow cytometry, then chemotherapy will be applied. To assess cell viability, MTT technique will be performed to evaluate apoptosis and flow cytometry technique with specific antibodies will be utilized. After, mRNA and protein of CD44, CD133, CD117 and ALDH1 genes will be evaluated by Real Time and ELISA, respectively. All results will be evaluated by appropriate statistical analysis. Studies of clinical response are needed to obtain a combination of molecular parameters prognostic factors. Hopefully, the results of this proposal contribute to new therapeutic strategies with information about estimation of prognosis and treatment decision (AU)
