Abstract

The heart corresponds to main target of TH and many of the classic signs and symptoms of hyperthyroidism are the cardiac and hemodynamic manifestations which include heart rhythm disorders. The present study has as aim to define the role of Caveolin-1 (Cav-1) depletion and oxidative stress in the pro-arrhythmic effects of hyperthyroidism. We will define whether Cav-1 deletion in mice leads to arrhythmias, will define in vitro whether cardiac myocytes exposed to high levels of thyroid hormone lose Cav-1 expression and by what mechanisms and integrate both approaches by examining whether intervening on the Cav-1/c-Src pathway can preserve adequate cardiac function in mice despite of elevated thyroid hormone levels. This last piece of the work being of very high clinical relevance. (AU)
