Abstract

Scientific discoveries have demonstrated the differentiation potential of stem cells from mesenchymal origin and its therapeutic application in both complex illnesses or injuries difficult to repair as in gene therapy. The aim of this study is to isolate, cultivate and characterize progenitor stromal cells of endometrial tissue from cows in phases II and III of corpus luteum formation. Additionally, the quantification of cytokine production by stromal progenitor cells under stimulation of LPS will be held, as well as the evaluation of viability pre and post-cryopreservation with two mediums of cryopreservation. For this purpose, 12 samples of endometrial tissue obtained from a slaughterhouse will be used, with 6 animals in the luteal phase II (n = 6) and 6 animals in the luteal phase III (n = 6). After harvest, the material will be properly manipulated in the laboratory to LANCA for isolation, culture and expansion in vitro of stromal cells. Later, it will be done the immunophenotypically characterization of cultured cells and also the differentiation for the tri- lineages. Also, cells will be stimulated with LPS for evaluating the immunomodulatory profile. Finally, the cells will be cryopreserved in two mediums of different cryopreservation and assessed for viability after one month of freezing. This experiment will provide data retrieval harvesting, cultivation, characterization, immunomodulatory profile and viability after cryopreservation of endometrial stromal cells from the tissue of cattle in view of the scarcity of studies on this species. Additionally, the data can provide a better understanding for the study of the pathophysiology of uterine disease in cattle, as well as provide better knowledge of the therapeutic application of stromal progenitor cells against these diseases, and the possible creation of cell banks for use and future studies.
