Influence of the amount of dentifrice and fluoride concentration on intraoral fluoride retention in children

Abstract

This study will evaluate fluoride (F) concentrations in saliva of children after brushing with dentifrices containing different F concentrations (0, 550 and 1100 ppm F) in different amounts (0,1, 0,3 and 0,5). Volunteers (n = 27, 8-10 years old) will be randomly divided into 9 experimental groups, according to the possible combinations of F concentrations in the dentifrices and amounts of the product to be used. They will be instructed to brush their teeth with a placebo dentifrice, 3 times/day, during one week (wash out period). On the 7th day, following a double-blind, crossover protocol, stimulated saliva will be collected prior to tooth brushing (baseline sample) with the assigned combination of dentifrice type and amount. Following, saliva samples will be collected at 5, 15, 30, 60 and 120 min after tooth brushing and the protocol will be then repeated for 8 additional weeks, in order to follow the crossover protocol. F analysis will be performed using the under oil inverted electrode, after buffering centrifuged samples with TISAB III. Data will be analyzed by ANOVA and Bonferroni's test. The significance of 5% will be selected a priori.
