Abstract

First characterized as a tumor suppressor, the role of PTEN (phosphatase and tensin homolog deleted on chromosome 10) on other cellular processes - such as proliferation, migration and survival - has been assessed more recently, showing a potential modulatory effect over neurogenesis and synaptic plasticity.The hippocampus is an important region for cognition - with a crucial function on learning and memory -, and is one of the centers of adult neurogenesis. Many environmental factors, such as exercise, has been showed as modulators of neurogenesis, contributing to positive cognitive effects.Through a conditional PTEN knockout system it is possible to circumvent the embriological lethality of whole-body PTEN deletion, in addition to enable a tissue- and time-specific intervention, providing an interesting tool for the study of its physiological functions. Therefore, this study aims to further characterize the PTEN signaling pathways on hippocampus, evaluating their biochemical and functional aspects modulated by PTEN partial deletion, physical exercise, and the interaction of both interventions. Thus, the results of this study could contribute to a better understanding of the pathophysiological role of PTEN, supporting the development of more efficient interventions aimed to prevent and/or treat pathologies related to the central nervous system - such as tumors, neurodegenerative processes and autism spectrum disorders.
