Abstract

Approximately 15% of eukaryotes contain supernumerary B chromosomes. When present, B chromosomes frequently represent as much as 5% of the genome. Despite thousands of reports describing the distribution of supernumeraries in various taxa, a comprehensive theory for the origin, maintenance, and evolution of B chromosomes has not emerged. Here, we propose sequencing the complete genomes of different species (the plant Cestrum intermedium, the coleopteran beetle Dichotomius geminatus and fish species Astyanax mexicanus) with and without B chromosomes to identify DNA sequences on the B. B sequences will further be analyzed through quantitative polymerase chain reaction and in situ hybridization. Genome sequencing based on Illumina next generation sequencing, comparative analysis, gene annotation and mapping and identification of B chromosomes will be performed. The development of the present research proposal will allow the identification of genes present on B chromosomes of different species and their possible biological role. Furthermore, we will be able to test the hypothesis that cell cycle genes tend to accumulate independently on B chromosomes of different species. (AU)
