Antimicrobial activity and physicochemical properties of different pastes used as intracanal medication

Abstract

The main function of the intracanal medication in pulpar necrosis is the antimicrobial action counter micro-organism which survived to the biomechanical preparation of root canal system. The combination of Calcium Hydroxide paste with other drugs has been suggested in order to increase the antiseptic capacity of the medication. The aim of this study is to evaluate the pH, calcium release, solubility and antimicrobial action on biofilms of the pastes: calcium hydroxide + saline solution, calen, calen PMCC, calcium hydroxide + chlorexidine, diantibiotic and triantibiotic. Materials and methods: The measurement of pH and calcium release: each group, containing 10 acrylic teeth with canal prepared will be filled with the pastes in study and so they have the crown access sealed and so will be immersed in deionized water and after 3, 24, 72 and 168 hours, they will be removed from the flasks and put in a new flask. The pH and calcium of the water that they were immersed will be measured by a pH meter and by an atomic absorption spectrophotometry, respectively. Solubility test: To evaluate the solubility, 60 acrylic teeth, that had them standardized cavities filled with pastes, will be immersed in deionized water and time before and after it, will have them volumetric measurements, using Micro-CT images. Antimicrobial activity: Biofilm in situ and in vitro of Enterecoccus faecalis, will be induced on 30 blocks of bovine teeth. After this, the samples will be treated by the pastes for 7 days, and then, incubated in heart-brain infusion for 24 hours. The percentage of living cells will be measured by using LiveDead dye and confocal microscope. The data will be compared statistically. (AU)
