Diacerein effects on left ventricle remodeling in a CHF model

Abstract

The heart failure is a crucial public health issue with increasing incidence and prevalence. Despite the recent therapeutical advances, the congestive heart failure (CHF) mortality and morbidity remain high. The knowledge of its pathophysiology and the development of new therapies are required. The inflammatory cascade has a significant role on left ventricle (LV) remodeling in patients with CHF. The modulation of the inflammatory process is a relevant target for CHF treatment. The diacerein is an anthraquinone that can inhibit the inflammatory process. Objective: To evaluate the diacerein effects on LV remodeling process in a model of left ventricle pressure overload using a rat model. Methods: Male Wistar rats five weeks old will undergo transverse aortic arch constriction by cervicotomy. Then, two groups will be treated with 50 mg/kg/day for four weeks while the other two groups will be treated with placebo (saline solution). At the end of the treatment period, the animals will be assessed by hemodynamic profile using pressure-volume loop catheter, fibrosis content by histology, and morphometric by lung and heart weight analysis. We hypothesize the diacerein will decrease the left ventricle fibrosis content with a consequent better left ventricle remodeling.
