Abstract

Diabetes mellitus (DM), a chronic metabolic disease in which blood glucose levels are increased, is the sixth most common cause of death worldwide. Its treatment involves the use of oral hypoglycemic drugs and, in severe cases, daily subcutaneous administration of insulin (INS), as it is an invasive route often leads to non-adherence of the patient to the treatment. In this sense, different strategies have been evaluated in order to find an alternative to INS administration, and the oral route is considered most convenient, safe and natural. However, oral administration of protein drugs is a major problem, mainly due to enzymatic degradation suffered in the upper portions of the gastrointestinal tract and low permeability across the intestinal mucosa. Thus, the encapsulation of INS in gellan microcapsules coated with resistant starch/pectin should promote the INS protection against degradation in the upper portions of the GI tract, allowing the release only the end portion of the bowel (colon specific release) and providing a contact closer to the intestinal mucosa, which should favor the permeation. During the research stage at the Faculty of Pharmacy (University D'Avergne) in Clermont-Ferrand, France, under the supervision of Prof. Dr. Eric Beyssac, gellan microcapsules coated with resistant starch / pectin will be assessed for its protective effect against the enzymatic degradation of INS and for its impact on increasing the bioavailability through permeability studies in Caco-2 cells.
