Abstract

The sugarcane has been widely used in production systems in the current scenario of Brazilian agriculture, with considerable importance in the economy and development of the country. However the massive expansion of culture throughout Brazil leads the need to develop improved cultivars and adapted to different climate conditions to which they are subjected. Biotic and abiotic stresses are factors that affect the productivity of a culture, and between these water stress is of great importance due to the rainfall and the impending temperature increase in the coming years. Analyze the expression of genes in plants under stress may contribute significantly to elucidate the routes of plant defenses, greatly contributing to the improvement of culture and the development of new varieties. The project aims to evaluate the functionality of some genes and their influence on the tolerance to water stress, including: SAM (S-adenosylmethionine decarboxylase); Pyruvato kinase; calmodulin; Lipid transfer protein; And UBQ-CoA reductase Cinnamoyl (CCR). Initially will be done a complete selection of ESTs from leaf library, generated by the Genome Project SUCEST, with further analysis in databases (NCBI and database Genome Project cane sugar SUCEST) and the acquisition of Storage Center genes (BCCCenter). The clones will be re-sequenced to prove its identity. The analysis of gene function is made by genetic transformation of Nicotiana tabacum via Agrobacterium tumefaciens, and then subjected to drought stress bioassay. Analysis of the expression of transgenes is confirmed by Real Time PCR. (AU)
