Abstract The reduction of the viable bacterial load after endodontic disinfectionprocedures is a key element for treatment success. Since ribosomal RNA(rRNA) indicates microbial viability, molecular approaches based on rRNA arechoice methods to assess the antimicrobial effectiveness of endodonticprocedures. The objective of the present study is to assess the viability of asyet-uncultivated bacteria after chemo-mechanical procedures duringendodontic retreatment using rRNA-based molecular methods. Samples from18 teeth with persistent/ secondary intra-radicular infection before (S1) and after(S2) chemo-mechanical preparation will be used in this study. cDNA, which wassynthesized from RNA, will be subjected to Real Time Polymerase ChainReaction (qPCR) with primers complementary for Bacteroidetes oral clone X083e Olsenella uli 16S rRNA sequences. Bacterial reduction will be analyzed bystatistical tests. | |
