Abstract

Tissue bioengineering is an emerging field that seeks alternatives to replace lost tissues and/or treatment of organs that went to failure. It uses biomaterials derived from biological tissues, synthetic or semi-synthetic, that mimic the extracellular matrix, associated or not with stem cells and/or bioactive molecules to promote tissue regeneration. The use of decellularized matrices as biomaterials (scaffolds) presents several advantages, such as chemical similarity to the host tissue, the three-dimensional architectural similarity; moreover, depending on processing, triggers a minimal immune response, decreasing the chances of rejection. The placenta has an extremely rich extracellular matrix. This, added to the fact that the placentas are usually discarded, make it a good alternative source of extracellular matrix to be used as biomaterials. Thus, the objective is to analyze the influence of freezing temperatures in the effectiveness of cell removal and maintenance of 3D matrix architecture. The collected placentas will be kept at different temperatures before the decellularization process. Histological analyses will be done by blades stained with Hematoxylin and Eosin, Masson's Trichrome and Picrosirius; architectural analysis of the matrix will be made by scanning electron microscopy; and the analysis of remaining DNA in the matrix will be held with PicoGreen assay.
