Abstract

The aim of this study will be to evaluate the expression IL1-B e TNF-a on tissue response to MTA in diabetic animals. Twenty rats will be arranged into two groups of ten animals each: group 1: normal rats, group 2: diabetics rats induced by Alloxan. All animals will receive two implants of polyethylene tubes, one containing sealer to be tested and an empty tube for control. After 30 days, the animals will be killed and the tubes with surrounding tissues removed and fixed. The tubes will be cut in half and a portion processed and included in glycol methacrylate to be observed in HE. Tissue reactions on the opening of the tube in contact with the material will receive scores as 0, none or few inflammatory cells and no reaction; 1, less than 25 cells and mild reaction; 2, between 25 and 125 cells and moderate reaction; and 3, 125 and more cells and severe reaction. The other half of the tubes will be placed in paraffin for immunohistochemical analysis for the presence of IL1-B and TNF-a. The immunostaining evaluation criteria will be: 1- high (3/4 of immunoreactive (IR) cells per microscopic field), 2- moderate (1/2 IR cells), 3 - low (1/4 cell IR) and 4 - null (absence of IR cells). The results will be analyzed by ANOVA and Kruskal Wallis tests (p<0,05).
