Analysis of the protein triad RANK, RANKL and OPG released after application of orthopedic forces in rats subjected to inhalation od cigarette smoke

Abstract

The habit of smoking is quite common and, most often, begins in adolescence. Among the systemic changes caused, the commitment with the cellular repair process has a great importance in understanding the dental- skeletal movements. There is no scientific evidence correlating this habit and its influence on orthodontic/orthopedic treatment, especially with the cellular and molecular analyzes. Objective: The objective of this study is to evaluate the process of bone remodeling after installation of orthopedic forces in smoking rats .Material and Methods: A total of 60 Wistar rats will be divided into 4 groups (n = 15): negative control [intact suture without cigarette smoke (CS)], experimental group I (intact suture and CS), experimental group II [rapid maxillary expansion (RME) and without CS] and experimental group III (RME + CS). Each group will be subdivided into 3 experimental periods of evaluation (n = 5) and the animals of each subgroup will be sacrificed after 3, 7 and 10 days after the orthopedic maxillary expansion. The changes in gene and protein expression of RANK, RANK-L, osteoprotegerin (OPG) will be evaluated. The PCR in real time will be used to amplify the cDNA genes of study, which results will be analyzed in the Sequence Detection Software vl.3. These datas will be confirmed with the evaluation of protein expression through Western Blotting. The results will be grouped according to the different studying groups and the experimental period, submitted to Analysis of variance (ANOVA) and compared by Student's t test. The difference will be statistically considered when p <0.05.
