Abstract

This study aims to evaluate the surface roughness of two new restorative materials and the formation of monotypic and heterotypic biofilms, as well as the viability of human gingival fibroblasts (FMM-1) in contact with these materials. For this, 63 blocks (4.5x4.5x1.5 mm) of each ceramic (high translucency zirconia partially stabilized by yttrium and lithium silicate reinforced by zirconium) will be made. The blocks will be standardized after polishing and will receive a glaze application. The surface roughness (Ra, Rz, RSm) will be evaluated through a profilometer and surface free energy will be determined by a goniometer. After sterilization, specimens will be contaminated for monotypic biofilm formation of S. mutans (ATCC 35688), S. sanguinis (ATCC 7073) and C. albicans (ATCC 18804) and for heterotypic biofilms with these microorganisms during 16 h formation. The biofilm formed on the materials will be quantified by counting the colony forming units (CFU/mL) and flow cytometry, and the viability of human gingival fibroblasts to the materials will be evaluated by MTT assay. Qualitative analysis of specimens will be performed by scanning electron microscopy, atomic force microscopy and confocal laser scanning microscopy. The results will be statistically analyzed and discussed against the literature. (AU)
