Abstract

The project deals the development of new Gd-based relaxation agents, taking account of the fine tuning of a number of parameters, namely improved sensitivity, very high stability in vivo, well-controlled excretion process and well-defined distribution pathway. The aim of the project will be to investigate the relaxometric behaviour of a series of new synthesized contrast agents based on the use of Gd-AAZTA functionalized with a short (C2 spaced) acid function (Gd-AAZTAC2COOH) which enables a further conjugation with lipophilic chains or multimeric species. The use of a short linker between the metal coordination cage and the functionalizing group should reduce the internal mobility of the paramagnetic core thus allowing the obtainment of high relaxivity systems. The contrast agents will be modified in order to address two important issues: i) the probes will be endowed by targeting capability through the conjugation to suitable vectors and ii) signal amplification (multimeric systems, nanosized carriers) will be provided in order to overcome the MRI low sensitivity threshold.
