Effects of genetic polimorphisms in health parameters in older women

Abstract

INTRODUCTION: The population ageing is a marked world phenomena. According to Papaléo Netto (1996), aging is a dynamic and progressive process, in which there are many morphological, functional, biochemical and psychological changes. Besides environmental factors related to life style, such as physical activity practice and feeding, genetics has a large importance to slow or to accelerate the ageing process because it contributes to explain the wide variability in its physiological and morphological declines. It is important to study the relationships among genetic polymorphisms, aging and environment to understand to phenotypes related to this phase of life. Polymorphisms are genetic variants in which the minor allele frequency is greater than 1% and many studies point out that genetic variants in the genes that codify AKT1, AGTR1 and visfatin can potentially explain the difference of magnitude in the aforementioned declines related to aging. Finally, studies that aim to study the relationships among aging, genetics and health parameters present large practice application because their findings will enable the prediction of health-related variables that will be more impaired in each person, which may help him/her to plan specific interventions to prevent theses health declines. OBJETIVES: To study the effects of AKT1 G205T (s1130214), AGTR1 A1166C (rs5186) and visfatin (rs4730153) polymorphisms in health parameters in older women. MATERIAL AND METHODS: This is a descriptive and cross-sectional study. Its sample will be composed by older women. Sample size will be calculated based in a pilot sample (10 for each group). The individuals will be shared in two groups for each polymorphism (AKT1: GG/GT or TT; AGTR1: AA/AC or CC; visfatin: GG or AG). Besides genotyping, the following evaluations will be done: anthropometric (body mass, stature and waist circumference), glycemia, lipidic profile, blood pressure, physical activity level (IPAQ), functional capacity (lower and upper extremity muscle strength, agility and balance, cardiorespiratory capacity and flexibility) and nutritional status (Formulário de Marcadores do Consumo Alimentar do Ministério da Saúde). Related to statistical analysis, it will be performed exploratory and descriptive analyzes, the data distribution will be analysed by Kolmogorov-Smirnov test and comparison between groups with different genotypes for each genetic polymorphism will be performed by independent samples t-test if the data are normally distributed (this is expected based on the scientific literature), using a significance level of 5%.
