Abstract

The B chromosomes are dispensable components that occur in addition to the standard complement (A) and exhibit non-mendelian heritage. Basically they are composed by repetitive DNAs and heterochromatic, more specifically by multigene families, transposable elements and satellite DNAs, and occur in about 15% of eukaryotes. Among grasshoppers, studies related to molecular composition and evolution of B chromosomes still scarce, being concentrated in some species such as Eyprepocnemis plorans and Locusta migratoria. Aiming to contribute to the knowledge of the evolutionary mechanisms acting in the diversification of repetitive DNAs of B chromosomes in grasshoppers, in this work it will be isolated and characterized satellites DNAs of the South American grasshopper Abracris flavolineata. This species presents a B chromosome in individuals occurring in the region of Rio Claro/SP. Additionally, we will also use the product of the B chromosome previously microdissected to amplification through PCR of the isolated satellite DNAs, aiming to compare the patterns of molecular evolution of these sequences between the A complement and B chromosome. Furthermore, the RNA of different tissues, developmental stages and with the presence of B chromosome of male and female specimens will be extracted to obtain the cDNA which will be used as template in order to check their levels of transcription of the sequences isolated. These analyzes will allow a deeper knowledge related on the genomic structure and evolutionary mechanisms operating in karyotype and supernumerary chromosomes of A. flavolineata. (AU)
