Abstract

The development of new treatments for diseases related to mammary gland depends on knowledge of morphology, physiology and breast pathology. The gerbils (Meriones unguiculatus) have been widely used in scientific experiments, since they are relatively easy to handle, robust and have small costs. This project aims to characterize morphologically normal mammary gland of Mongolian gerbils and to evaluate the carcinogenic effect of bisphenol A by morphological and immunohistochemical analysis to determine early-tumoral lesions. For the step of morphological characterization, animals in puberty, pregnant, in lactation and senile will be used. The mammary glandular tissue will be stained with hematoxylin and eosin, Gomori reticulin and PAS. Also, immunohistochemical reactions for ER-alfa, ER-beta and progesterone receptors and alfa-actin will be performed. For the carcinogenic effect of bisfenol A in the mammary gland, the animals will be treated during pregnancy and lactation with different doses of bisphenol A or estrogen. From the females born, one group will be treated with carcinogen N-nitroso-N-methyl-urea and the other group will be maintained as control. The pre-neoplastic cell damage will be assessed by staining with hematoxylin and eosin, PAS and the immunohistochemical reactions for the presence of Ki67 and PCNA antigens and ER-alfa, ER-beta and progesterone receptors. In addition, we will analyze other organs for the presence of metastases. At the time of euthanasia, blood will also be collected for serum estradiol dosage.
