Accuracy of the ultrasonography (Mode-B and Doppler) in the determination of superovulatory response in ewes

Abstract

The present study aims to verify the accuracy in determining the superovulatory response of sheep by sonographic evaluation (Mode-B and Doppler). For that, twenty-four Santa Inês ewes will be subjected to superovulation protocol. In a random estrous cycle of females (Day 0), estrus synchronization is performed by inserting an intravaginal progesterone device (CIDR®) that will remain until Day 8. At the same time the insertion and removal of CIDR device (Days 0 to 8), equivalent doses (37,5¼g PGF± analog, D-cloprostenol - Prolise®) are i.m. administered. The superovulation treatment will start 48 hours before the removal of CIDR device (D6), while females receive different total doses of exogenous porcine FSH (200, 133 or 100 mg of FSH). Total doses will be administered in eight applications at 12 hour intervals (20, 20, 15, 15, 10, 10, 5 and 5% FSHp). On Day 6 (1st application of FSH), the females will also receive i.m. injection of 300 IU of eCG (Novormon®). On the corresponding day of the embryo collection, the females will be evaluated by ultrasonography (Mode-B and Doppler) for the purpose of identifying and quantifying corpora lutea and anovulatory follicles (diameter e 5 mm) present in the ovaries. Following, the females will be submitted to laparoscopy with the aim of evaluating the same ovarian structures, which will be quantified for later correlation with the ultrasound findings, using the Pearson correlation test using SAS software.
