Abstract

Considering the national importance of Mangalarga Marchador (MM) breed and its increasing internationalization, studies evaluating different aspects of their unique gaits - marcha batida (MB) and marcha picada (MP) - are needed. The objectives of this study are to determine the kinematic variables (VCine) of MB and MP of previously awarded horses; to verify associations between morphometric measurements and VCine; to verify the differences in VCine between MM horses with homozygous and heterozygous genotypes for the DMRT3 gene for each type of gait; and to identify, through large genome-wide association study (GWAS), SNPs and genomic regions containing possible genes responsible for the phenotype of MB in MM breed. 12 horses of each gender and each gait (total 48 horses) will have 35 morphometric measurements assessed and 23 VCine determined by three-dimensional system. The MB and the MP will be compared by unpaired Student's t test (Pd0.05) and associations between morphometry and kinematics will be evaluated by Pearson correlation. The SNP g.22999655C>A of DMRT3 equine gene will be analyzed by genotyping protocol by PCR-RFLP technique and correlated with VCine. The DNA samples from each individual will be genotyped with Equine SNP70 BeadChip. Association study will be performed using the Cochran-Armitage test with 1 degree of freedom implemented by Plink software v1.07. Positions of SNPs and genes will be considered based on the latest assembly sequence of the horse genome (EquCab2.0 available on the National Center database for Biotechnology Information -NCBI).
