Abstract The installation of osseointegrated implants in patients under bisphosphonates treatment in controversial. Recent studies demonstrated that patients under this treatment can show diverse clinical outcomes after implants installation, that can vary from better osseointegration to lost of implants. However, few is known about the factors that can be involved on the success of this approach on bisphosphonate-treated patients. The aim of this study will be to evaluate the effect of two different bisphosphonates, Zoledronic Acid and Sodium Alendronate on osteoblasts cultured onto titanium surfaces. Machined titanium disks will be inserted on cell culture plates in addition to culture medium containing selected concentrations of bisphosphonates (0 - control, 0,1, 0,5, 1 and 5¼M), for 48 hours. After this period, human osteoblasts (SaOs-2 ATCC #HTB85) will be seeded on the titanium surfaces. After 7, 14 and 21 days cell adhesion by scanning electronic microscopy, cell viability, total protein production, alkaline phosphatase activity, collagen synthesis and mineral nodule formation will be evaluated. Data obtained will be analyzed by selected statistical tests, according to normal curve adherence and homocedasticity at 5% significance level. | |
