Apocynin: an antioxidant or a NAD(P)H oxidase inhibitor in vascular cells?

Abstract

Reactive oxygen species (ROS) play important roles in the pathogenesis of cardiovascular disorders, such as hypertension. The isozymes of NAD(P)H oxidases are source of ROS in the organism, and increased activity of them has been associated to the hypertension. The development of inhibitors of NAD(P)H oxidase and antioxidant drugs is recommended for the treatment of hypertension. In this project, we will compare the effect of apocynin to the different chemical compounds, such as protocatechuic acid and ellagic acid, having antioxidant effect and NAD(P)H oxidase inhibitory effect, respectively. The vasodilatory effects, and the effects of these compounds on the contractions induced by ±-1 adrenergic agonist, will be assessed by functional experiments in aortas. The antioxidant activity of the compounds, the effect of the compounds on lipid peroxidation and NAD(P)H oxidase- dependent ROS production,will be evaluated by the FRAP method, the TBARS method, and lucigenin chemiluminescence, respectively. The results will be compared among the compounds employing the most appropriate statistical test (ANOVA or Student's t) and the differences will be considered significant when p <0.05.
