Objective sleep evaluation and mean telomere length in a sample of São Paulo city

Abstract

Sleep is one of the most important physiological processes for the healthy maintenance of many organisms. Significant alterations in sleep patterns and the quantity and quality of sleep are found as people get older. Besides, the elderly population has been found to have a higher prevalence of sleep disturbances. Concurrently with increasing age, there is also the shortening of telomeres, structures located in the terminal region of chromosomes, as a result of cellular senescence mechanisms. Telomere shortening has been investigated as a marker of biological aging and age-related conditions. Taking into consideration the simultaneous occurrence of telomere shortening and changes in sleep structure with aging, the present proposal aims to measure the mean telomere length of leukocytes in a sample from EPISONO project, a single-center study that described the prevalence of the main sleep disorders in a representative adult population (20 to 80 years) sample of São Paulo city. For the present project, DNA collected and extracted from peripheral blood of 1042 individuals from EPISONO will be used. Mean telomere length of leukocytes will be measured by quantitative real-time polymerase chain reaction method, using only one fluorophore (MMqPCR). It will be verified the association between relative telomere length and socio-demographic variables, objective and subjective sleep parameters e the presence of sleep disorders. Thus, the project aims to contribute to the identification of potential markers that may assist in the diagnosis or help identify therapeutic targets of sleep disorders or related phenotypes in this representative sample of the population of São Paulo.
