Abstract

The beetle larvae Sphenophorus levis are one of the major pests of sugarcane plantations in the state of São Paulo, and so far, no method of control has been effective against the insect. Within this context, control measures are necessary to minimize the damage caused by this pest. This project envisages in the development of transgenic sugarcane more resistant to Sphenophorus levis. Initially, the transcriptome data of S. levis obtained from studies at the Laboratory of Molecular Biology, Department of Genetics and Evolution, Federal University of São Carlos, would be analyzed to identify target genes for gene silencing by RNAi. The objectives of the study are to identify target genes from the transcriptome data, develop vector constructs with target genes to be silenced, to generate transgenic sugarcane through both Biolistics and Agrobacterium mediated transformation methods and to evaluate the transgenics for S. levis resistance. The targets preliminarily studied at the laboratory which includes an encoder of a serpin and a V-ATPase gene will also be used for sugarcane genetic transformation.
