Study about the cellular transport of exosome subunit Rrp44 in Saccharomyces cerevisiae

Abstract

Saccharomyces cerevisiae is an yeast pretty important for the currently society, by being a model of studies and being part of huge industrial productions. This yeast has a processing RNA machinery called exosome, which has two main role: mRNAs degradation and 5,8S rRNA processing. The exosome has a RNase PH-like ring, which one is composed by the following subunits: Rrp41-Rrp45-Rrp43-Rrp46-Rrp42-Mtr3. Besides that, there are 3 more subunits (Rrp4Rrp4, Rrp40 e Csl4) that together with the others ones build the core. The citoplasmatic exosome found in yeast has 10 subunits, including the Rrp44 which interacts with the complex. The nuclear exosome has 11 subunits, adding the subunit Rrp6. Nowadays, the scientific community supposes that the Rrp6 is transported to the nucleus by its nuclear location sign (NLS), however the process of how the others subunits go to the nucleus still unclear. This project aims to analize the nuclear transport of the S. cerevisiaes exosome subunit Rrp44p.
