Abstract

Extracellular matrix (ECM)-signaling is crucial for determination of cell fate and behavior, during mammary gland morphogenesis and homeostasis. However, little is known about the molecular mechanisms that regulate these processes. The Hippo pathway, a cascade involved in the regulation of several cellular processes, including organ size, seems to be an important candidate to as a mediator of ECM signals that change gene expression programs. Our laboratory has been exploring the hypothesis that Hippo is involved in crucial processes of mammary gland development and biology and it can intermediate the signals of the ECM that regulate these processes. Our preliminary results indicate that the tissue architecture and the basement membrane, an ECM component of epithelia and other tissues, influence the location, concentration and activity of YAP, an effector of the Hippo pathway. In this context, the goal of this proposal is to identify the proteins that interact with YAP in mammary epithelial cells in response to the basement membrane. We will use proximity-dependent biotin identification (BioID) followed by mass spectrometry analysis. We expect that the identification of YAP molecular partners will reveal an interaction network that may bring crucial information on the ECM-signaling mechanisms during mammary gland morphogenesis and homeostasis.
