L. theobromae biomass: culture medium nutrient or inoculum in new batch, aiming EPS production at different temperatures

Abstract

The production of exopolysaccharide (EPS) by filamentous fungus Lasiodiplodia theobromae has industrial interest, due to its potential application related to its immunomodulatory action, inducer of pro-inflammatory and anti-tumor response and anticoagulant action when sulfated. However, the industrial viability of this bioprocess still needs to overcome challenges, such as cell growth 10 times higher than the production of EPS, generating a large content of residue in the form of biomass and low productivity (carbon source concentration / EPS concentration). This project is based on previous studies in which was verified: 1) production of EPS medium using the lyophilized biomass from previous culture in a new batch of culture with L. theobromae; 2) cell growth and secretion of jasmonic acid when the L. theobromae biomass, after maximum EPS production, was aseptically transferred to new culture medium and condition. Thus, this work has as objective using strategies to optimize EPS production by L. theobromae, analyzing the influence of different culture temperatures (between 25-40°C), using both the lyophilized biomass as nutrient and alive and viable biomass as inoculum for a new production batch.
