Abstract

The embryoid polarity of the embryo has a fundamental role in the embryonic axis determination, in a way that it generates a differential signalling in embryo regions during its development, allowing the definition of different cell populations. The definition of those axes depends on several signalling pathways. One of those is the Wnt/²-catenin signalling pathway, which is essential to the anteroposterior axis definition in early embryos, when this pathway is active it defines a posterior region and when it is inactive it defines an anterior region. This polarity has also been seen in embryo study models, the embryoid bodies, which are cellular aggregates from embryonic stem cells, capable of reproducing events of the early embryonic development and can give rise to cells from the three germ layers. Previous studies, our laboratory suggested the role of the COUP-TFII (Chicken Ovoalbumin Promoter-transcription Factor II) during the acquisition of cellular fates and observed a polarized expression of this gene in embryoid bodies. The idea of the polarized Wnt/²-catenin together with the polarized expression of COUP-TFII, made us wonder whether there is a relationship between those two elements. In this study we will create a cell lineage from a murine embryonic stem cell lineage (E14TG2a) expressing the Green Fluorescent Protein (GFP) under the control of the Wnt/²-catenin pathway, then we will put them under the differentiation process by generating embryoid bodies with those cells. Finally, the analysis will be done by immunohistochemistry to visualize the expression pattern of COUP-TFII along with the expression pattern of the Wnt/²-catenin pathway. (AU)
